藏茶水提物对高糖脂饮食小鼠血脂及其肠道菌群的影响分析

摘要:

为探究雅安藏茶水提取物对高糖脂饮食小鼠的血脂调节及肠道菌群的影响,将18只6周龄SPF级雄性昆明小鼠随机分为3组,其中对照组饲喂基础饲料,模型组和干预组小鼠饲喂高糖脂饲料。饲喂12周后,干预组按照0.2 mL/10(g·bw·d)的剂量灌胃藏茶水提取物,模型组和对照组同时灌胃相同体积的生理盐水。灌胃14周后,检测各组小鼠血脂水平,分析小鼠肠道菌群的变化。结果显示,与对照组相比模型组小鼠血清中TC、LDL-C含量显著增高(P<0.01);与模型组相比干预组血清中TC、TG含量明显降低且具显著性差异(P<0.05),LDL-C含量虽无显著性差异(P>0.05),但有一定的下降趋势,而HDL-C无显著性差异(P>0.05)。同时,雅安藏茶能够有效降低高糖脂饮食小鼠的血糖。菌群分析表明,模型组小鼠肠道菌群结构发生紊乱,而干预组有益菌属如norank_f_erysipelotricaceaeLactobacillus等的丰度有所增加,有害菌属如 [Ruminococcus]_torques_group、 Clostridiaceae_1等的丰度有所下降。同时,干预组菌群整体结构趋同于对照组,与模型组有显著差异。由此可见,高糖脂饮食对小鼠肠道菌群造成紊乱,而藏茶水提物可有效改善其血脂水平和肠道菌群结构。


Effects of Tibetan tea water extract on blood lipids and gut flora in mice fed with high sugar and fat diet

Abstract: In order to explore the effect of Ya’an Tibetan tea water extract on blood lipid regulation and gut flora in mice induced by high-sugar and high-fat diet, eighteen 6-week-old SPF male Kunming mice were randomly divided into three groups. The control group was fed with basal feed, while the model group and the intervention group were fed with high-sugar and high-fat feed. After 12 weeks of feeding, the intervention group was given 0.2 mL/10 (g·bw·d) of Tibetan tea extract, while the model group and the control group were given the same volume of normal saline. After 14 weeks of treatment, the blood lipid levels of the mice in each group were detected, and the changes in the gut flora of the mice were analyzed. The results showed that compared with the control group, the serum levels of TC and LDL-C in the model group were significantly increased (P<0.01); compared with the model group, the serum levels of TC and TG in the intervention group were significantly decreased (P<0.05), although there was no significant difference in LDL-C content (P>0.05), there was a certain downward trend, while there was no significant difference in HDL-C (P>0.05). At the same time, Ya’an Tibetan tea can effectively reduce the blood glucose of mice in high glycemic and lipid diet. The microbiota analysis showed that the gut microflora structure of the mice in the model group was disordered, while the abundance of beneficial bacteria such as norank_f_erysipelotricaceae and Lactobacillus increased and the abundance of harmful bacteria such as [Ruminococcus]_torques_group and Clostridiaceae_1 decreased in the intervention group. At the same time, the overall structure of the microflora in the intervention group was similar to that in the control group, which was significantly different from that in the model group. In conclusion, the gut microflora of mice is disturbed by high carbohydrate and lipid diet, and tea extract can effectively improve the level of blood lipid and the structure of intestinal microflora.
