摘要:

建立超高效液相色谱-四极杆飞行时间串联质谱检测有柄石韦抑菌成分的方法,用乙酸乙酯和水获取有柄石韦粗提物,Intersustain C18色谱柱、流动相A乙腈溶液和流动相B 0.1%甲酸水溶液等为色谱条件,正、负离子质量扫描范围均为50~1000 ,应用UPLC-XevoTM TQ MS进行物质鉴定。采用倍比稀释法进行山奈酚与圣草酚对金黄色葡萄球菌的MIC和MBC测定。在柄石韦乙酸乙酯提取物中鉴定出19种化合物,其中山奈酚为高含量化合物之一,水提取物中鉴定出17种化合物,圣草酚为高含量化合物之一。山奈酚对S. aureus的MIC和MBC值分别是0.625 mg/mL和1.25 mg/mL,圣草酚分别是0.25 mg/mL和0.50 mg/mL。该UPLC/Q-TOF/MS法灵敏、准确、通量化,能为天然产物分析与应用提供研究基础。


Detection of antimicrobial extracts of P. petiolosa by UPLC/Q-TOF/MS

Abstract: Objective to establish a method for the determination of antibacterial components in P. petiolosa by ultra performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF/MS). Ethyl acetate and water were used to extract the crude extract of P. petiolosa. Intersustain C18 column, mobile phase A acetonitrile, mobile phase B 0.1% formic acid and so on were the chromatographic conditions. The scanning range of positive and negative ions is 50-1000  on UPLC-XevoTM TQ MS. The MIC and MBC of kaempferol and eriodictyol against S. aureus were determined by multiple dilution method. The 19 compounds were identified from the ethyl acetate extract of P. petiolosa, among which kaempferol was one of the high content compounds. The 17 compounds were identified in the water extracts, among which eriodictyol was one of the high content compounds. The MIC and MBC values of kaempferol against S. aureus were 0.625 mg/mL and 1.25 mg/mL, respectively. The corresponding two values of eriodictyol were 0.25 mg/mL and 0.50 mg/mL. UPLC/Q-TOF/MS method has the advantages of sensitivity, accuracy and flux, which can provide research basis for the analysis and application of natural products.
