摘要:

基于偏最小二乘回归分析法(PLS)分析灵芝子实体体外抗肿瘤活性谱效关系。采用HPLC法建立灵芝药材指纹图谱,评价不同灵芝子实体对人肺腺癌细胞A549的细胞增殖抑制率,运用PLS法关联指纹图谱共有峰的相对峰面积与A549细胞增殖抑制IC50,建立谱效关系模型。结果表明:灵芝子实体指纹图谱中有14个共有峰,其中有6个共有峰的回归系数与A549细胞抑制IC50呈负相关,其贡献大小排序为X5(灵芝烯酸B)>X8(灵芝酸A)>X12(灵芝酸D)>X4X9(灵芝酸H)>X2(灵芝酸C2)。灵芝子实体体外抗肿瘤药效作用是多种成分共同作用的结果,可为灵芝药材质量控制提供参考。


Analysis of fingerprint and antitumor activity in vitor of Ganoderma lucidum by HPLC

Abstract: To explore the relationship between HPLC fingerprint and antitumor activity in vitro of the alcohol extraction of Ganoderma lucidum fruit bodies by PLS. The fingerprint of Ganoderma lucidum was established by HPLC. The inhibitory rate of A549 of Ganoderma lucidum was evaluated. PLS analysis was used to study the relationship between the HPLC fingerprint and the antitumor activity.  A total of 14 common substance peaks were identified, and 6 of them were negatively correlated with the Growth inhibition IC50 values of A549 cells. The contribution order is X5 (Gano11derenic acid B) > X8 (Ganoderic acid A) > X12 (Ganoderic acid D) > X4 > X9 (Ganoderic acid H) > X2 (Ganoderic acid C2). The antitumor activity of Ganoderma lucidum is the result of many components. This study can provide reference for quality control of the medicinal materials.
